For me this was one of the most interesting and moving scenes in RotK. It highlighted the strength of Frodo and Sam's friendship, their perserverance and determination to achieve their goal, no matter what it costs them personally. For me, humour at this point would be utterly out of context and ruin the whole scene/s- it's
meant to show a more serious side of the movie and thus would be ruined by any humour in that scene, as it is inappropriate for for that part of the movie. However, it's a personal feeling and people will obviously have differing opinions, which are all valid.
